Preparation

Sample preparation is an important aspect of electron microscopy. Biological specimen have to be fixed to be stable in the low-pressure environment of the microscope. For transmission electron microscopy, samples also have to be thinned to ~100 nm for electrons to pass through the sample. Sample preparation infrastructure includes instruments for carbon coating, plasma cleaning, high-pressure freezing, vitrification, freeze substitution and (cryo-) ultramicrotomy.

 

  • Emitech K950 Carbon coater and glow discharger

    The Emitech K950 Carbon coater with glow discharging unit is use for preparation of carbon foils as sample support and for glow-discharge preparation to hydrophilise EM grids.

  • Emitech K975 carbon coater

    The Emitech K975 is a carbon coater that is water-cooled and pump by a turbo to high vacuum. It has a unique constant pressure carbon gun for controlled evaporation and can perform sequential coatings without breaking vacuum. The K975 is able to measure carbon layer thickness during evaporation and is mainly used in the lab to prepare thin carbon layers of specified thickness.

  • Leica EM GP

    The Leica electron microscopy grid plunger (EM GP) is used to vitrify thin samples such as proteins, viruses, bacteria, and thin cells by plunging them into liquid ethane sucg that they are cryo-fixed in a thin layer of vitreous water. 

  • Leica EM Pact 2 High Pressure Freezer

    The Leica EM PACT 2 is a high-pressure freezer for vitrifying samples up to 200 um in thickness, like cell suspensions and tissues. It is equipped with a rapid transfer system for correlative light and electron microscopy studies.

  • Leica EM AFS

    The Leica EM automatic freeze substitution device automatically freeze substitutes the vitreous water in frozen embedded samples at low temperatures with plastic while doing chemical fixation to make samples suitable for room temperature sectioning.

  • Baltec CPD030 Critical Point Dryer

    The Baltec CPD 030 critical pojnt dryer prepares samples for SEM.

  • RMC ATUMtome

    The RMC ATUMtome can automatically section a fixed tissue block into many slices on a ribbon to be put on a wafer for imaging by SEM. Many slices can be combined into a tomogram to generate a large volume block.

  • Leica EM UC6 ultramicrotome

    The Leica EM UC6 is designed for ultrathin sectioning of biological and industrial samples. The ultramicrotome is equipped with a touch screen control panel, a programable knife, a Leica MZ6 stereomicroscope, a Leica IC A video module and an “ErgoWedge” which makes the UC6 adaptable to the operator’s height and position. 

  • Leica EM UC6 Ultramicrotome

    The Leica EM UC6 ultramicrotome with Leica FC6 cryochamber is suitable for cryo sectioning frozen hydrated materials.  It is contained in a nitrogen gas flooded dry chamber and a charger for firm attachment of cryo sections to EM grids.

  • Leica EM UC7 ultracryomicrotome

    The Leica EM UC FC7 is an ultramicrotome with cryochamberfor sectioning frozen or room temperature samples.
